Long-Range Cooperative Disassembly and Aging During Adenovirus Uncoating

نویسندگان

چکیده

The disassembly of a virus' protein shell must happen at the right time to ensure transfer viral genome its host. New experiments reveal that aging determines dynamics this unraveling.

DNA-Protein Cooperative Binding through Long-Range Elastic Coupling

Cooperativity plays an important role in the action of proteins bound to DNA. A simple, mechanical mechanism for cooperativity, in the form of a tension-mediated interaction between proteins bound to DNA at two different locations, is proposed. These proteins are not in direct physical contact. DNA segments intercalating bound proteins are modeled as a Worm-Like Chain, which is free to deform i...

Monitoring dynamics of human adenovirus disassembly induced by mechanical fatigue

The standard pathway for virus infection of eukaryotic cells requires disassembly of the viral shell to facilitate release of the viral genome into the host cell. Here we use mechanical fatigue, well below rupture strength, to induce stepwise disruption of individual human adenovirus particles under physiological conditions, and simultaneously monitor disassembly in real time. Our data show the...

Insights into Adenovirus Uncoating from Interactions with Integrins and Mediators of Host Immunity

Human adenoviruses are large (150 MDa) nonenveloped double-stranded DNA (dsDNA) viruses that cause acute respiratory, gastrointestinal and ocular infections. Despite these disease associations, adenovirus has aided basic and clinical research efforts through studies of its association with cells and as a target of host antiviral responses. This review highlights the knowledge of adenovirus disa...
